摘要
This seminar provides an introduction to the 10x Chromium System, which is capable of partitioning cells or HMW DNA into droplets, and preparing sequencing libraries in a highly parallel fashion such that all fragments produced in a single partition share a common barcode. The Chromium Single Cell applications allow both 3' gene expression profiling of hundreds to millions of cells, as well as the assembly of full-length V(D)J sequences on a cell-by-cell basis for detailed analysis of the adaptive immune system. On the DNA side, Chromium Genome kit employs Linked-Reads to fully resolve genetic phasing, reveal structural variation and detect variants in previously inaccessible and complex regions of the genome. The talk will give an overview of all three applications and some of the research discoveries they are being used to unlock.
